Major Research Progress

In order to search for naturally occurring bioactive metabolites from fungiwe investigated the chemical constituents of more than 200 fungi in Southwest of China.  A library contained more than 2000 including 700 novel compounds including terpenoids, phenolics and nitrogen-containing compounds from basidiomycetes and ascomycetes (Chem. Rev.2005, 105: 272; Chem. Rev. 2006, 106: 2209; Nat. Prod. Rep. 2010, 27: 1531; Nat. Prod. Rep. 2011, 28: 787) were established. Some of them showed very interesting pharmacological activities. Two drug candidates are evaluating for their potential as drugs before clinic.

Yunnan Sudden Deaths syndrome is responsible for more than 300 deaths in Yunnan Province, southwest China over the past 30 years. It has long been a mystery and a fascinating problem. We have identified the new toxic mushroom T. venenata, isolated two new toxic unusual amino acids from mushroom Trogia venenata guided by toxicity tests using mice, and detected directly new amino acid in a victim’s blood. Mice treated with both amino acids exhibited an increase in serum creatine kinase (CK) activity. These new toxins are the cause of ‘Yunnan Sudden Death’ syndrome, an important local epidemic of, as yet, undefined etiology (Angew. Chem. Int. Ed. 2013, 51: 2368;Science 2010, 329: 132; Science 2012, 335: 1293). We solve the problem. A campaign to warn people against eating the tiny mushroom with printed brochures has dramatically reduced the number of deaths, with none reported in 2010-2014! At least we have saved more than 80 people’s life.

Twelve people were hospitalized for severe weakness and muscle loss after eating the mushrooms, and three died. The poisoning the mushroom causes is referred to rhabdomyolysis (N. Engl. J. Med. 2001, 345: 798). We have identified the toxins found in a previously unknown poisonous European mushroom Tricholoma terreum. Fifteen novel triterpenoids were isolated from the fruiting bodies of the toxic mushroom T. terreum. Two abundant compounds in the mushroom displayed acute toxicity when administered orally in mice. Both compounds were found to increase serum creatine kinase levels in mice, indicating that T. terreum may be the cause of mushroom poisoning ultimately leading to rhabdomyolysis (Chem. Eur. J. 2014, 20: 1531).

Obesity represents a disorder of physiology whereby there is raised food intake resulting in increased deposition of fat within body tissues. There is an increasing incidence of obesity within the population.Obesity is associated with increased risk of diabetes, hypertension, cardiovascular disorders and renal diseases, which afflict 40% of these individuals. Clearly in both health and economic terms this represents a major treatment population. The lipase is one of the important target of anti-obesity drugs. Vibralactone, an unusual fused b-lactone type metabolite, was found to inhibit pancreatic lipase significantly. The structure and absolute configuration of vibralactone from the cultures of Basidiomycete Boreostereum vibrans was established by spectroscopic methods and computational methods. (Org. Lett. 2006, 8: 5749). Using vibralactone as original template the structure was optimized and more than 200 derivatives have been synthesized. We found a new compound with pancreatic lipase inhibitory activity with an IC50 in 14 nM which is 3000 times potent than vibralactone (applied patent). It has been as a drug candidate in evaluation. The unique biosynthetic pathway of vibralactone including several very interesting reactions which may involve unusual enzymes was elucidated (Angew. Chem. Int. Ed. 2013, 52: 2298).

We also discovered a series of new antitumor and anti-HIV agents from different fungi and clarified their mechanisms (Oncotarget 2014, on line; Cell Death Dis. 2013, 4: e804; Euro. J. Cancer 2011, 47: 316; Cancer Lett. 2007, 258: 199; Patent: US 7,659,308 B2; JP 4769726; Biochem. Biophys. Res. Comm. 2007, 355: 1091; Chemistry & Biodiverity 2008, 5: 447).
